STEAP1 has emerged as an ideal target in cancer therapeutics. However, the functions of STEAP1 in hepatocellular carcinoma (HCC) remain unexplored. In the current study, we sought to characterize the biological roles of STEAP1 in HCC. STEAP1 transcripts are over-expressed and associated with poor clinical outcomes in patients with HCC in several publicly available datasets. STEAP1 silencing using small interfering RNA inhibited cell growth and was accompanied by G1 arrest induced by the suppression of cyclin D1 and the promotion of p27. STEAP1 silencing suppressed the expression of c-Myc, which we identified as a component in STEAP1 signal transduction by mining publicly available datasets, and confirmed this by PCR array. In conclusion, the knockdown of STEAP1 in HCC cells led to cell-growth inhibition involving G1 arrest by targeting the suppression of c-Myc. This study suggests a preclinical concept for STEAP1 as a druggable target in HCC.

Metastatic / recurrent colorectal cancer is a disease with a poor prognosis, and the number of deaths continues to increase, and the development of more effective treatment methods is awaited. The FDG-PET examination, which utilizes the characteristics of increased sugar uptake in colorectal cancer, is useful for diagnosing colorectal cancer. Therefore, anticancer drugs using FDG as a carrier are promising as therapeutic agents for colorectal cancer. Therefore, we aimed to prepare an FDG-binding anticancer drug and examine its effect. A fluorescent reagent directly bound to FDG was prepared and examined, but no difference of anticancer effect from other sugar was observed. Considering that the stability of the reagent was a problem, we prepared a fluorescent reagent in which FDG was bound to liposomes, and found that it was taken up by cancer cells. In the future, we would like to promote the development of new anticancer agents that utilize this mechanism using FDG-binding liposomes.

Six-transmembrane epithelial antigen of the prostate 1 (STEAP1), identified in prostate cancer cells as a cell surface protein, is over-expressed in a subset of human cancers. In cases of Ewing sarcoma, STEAP1 augmented cell proliferation and invasion and was accompanied by increased reactive oxygen spices (ROS) levels. Here, we characterized the biological impact of STEAP1 and ROS in multiple myeloma (MM) pathogenesis. STEAP1 expression negatively correlated with OS as determined from a publicly accessible gene expression profile data set. A loss-of-function approach in cultured MM cell lines revealed that STEAP1 silencing suppressed migration, invasion and bone marrow homing through inhibition of ROS production. Mechanistically, the inhibition of STEAP1 was associated with increased expression of anti-oxidant molecules regulated by the transcription factor, nuclear erythroid 2-related factor (NRF2), in MM cells. This study identified STEAP1 as a therapeutic target for MM.

Multiple myeloma (MM) is characterized by clonal proliferation of long-lived plasma cells within the bone marrow. Despite recent advances in its treatment, MM remains an incurable disease, underlining the need to continue exploring its molecular characteristics. We have identified that BCL9 has a pivotal role in MM pathogenesis. In this study, we found that CYBRD1, which is a iron regulating molecule, as a novel therapeutic target in MM using BCL9-Chromatin Immunoprecipitation (ChIP)-Sequence (Seq) (ChIPSeq).

We developed a stabilized a helix of BCL9 (SAH-BCL9), which we show targets beta-catenin, dissociates native beta-catenin/BCL9 complexes, selectively suppresses Wnt transcription, and exhibits mechanism-based antitumor effects. SAH-BCL9also suppresses tumor growth, angiogenesis, invasion, and metastasis in mouse xenograft models of Colo320 colorectal carcinoma. By inhibiting the BCL9/beta-catenin interaction and selectively suppressing oncogenic Wnt transcription, SAH-BCL9 may serve as a prototypetherapeutic agent for cancers driven by deregulated Wnt signaling.

【目的】癌の治療戦略として注目されているがん間質に対する治療法を開発する目的で本研究を行った。【背景】がん随伴細胞(CAF)が癌細胞より分泌された種々の増殖因子により線維芽細胞より変態した筋線維芽細胞に類似したものであることから、肝星細胞と同様にVitamin Aを取り込む性質を持つことに着目した。 【方法および結果】平成20年度はVA-liposomeを作製し(Sato Y, Takimoto R, Nat Biotech 2008)、in vitroでのCAFへの導入効率を検討し、VAとリポゾームのratioなど効率的な送達を目指した。また、乳癌細胞、大腸癌細胞など各種癌細胞株とCAFの共培養系を確立し、抗がん剤を搭載したVA-リポゾームによる細胞障害性を検討した。これらのin vitroの結果では、VA-リポゾームはCAF特異的に抗がん剤を送達し、CAFの障害により間接的にがん細胞の増殖も抑制しうること示した。そのうえで、平成21年度はプロトタイプのVA-リポゾームを作製し、これに標識化合物(siRNA, plasmid DNAなど)を封入しin vivoで担癌マウスに尾静脈から投与し、in vivo image analyzerで解析した。その結果、CAFのみならずCAFを通じて癌細胞へも特異的に標識化合物を送達しうることを確認した。現在、これらの結果を踏まえて、抗がん剤を搭載したVA-リポゾームを開発し、抗腫瘍効果の増強による新しい細胞治療を目論んで準備を進めている。

非アルコール性脂肪性肝炎発症に関わる鉄代謝異常の分子機構解析と治療法の開発

本邦における非アルコール性脂肪肝(NAFLD)患者は約800万人にものぼると推定されている.NAFLDの中の約10%にあたる80万人が肝硬変に進展し,さらには肝細胞癌の発生母地となる非アルコール性脂肪性肝炎(NASH)であると考えられている.しかしながら,NASHの発症・進展の機序は依然不明であり,その治療法も確立されていないのが現状である.最近NASHの発症・進展には肝への鉄過剰蓄積によって惹起される酸化ストレスの関与が示唆されている.われわれの検討結果から,NASHにおいては肝臓に鉄が過剰に蓄積し,血清フェリチン値やトランスフェリン飽和度が上昇していることから,鉄がNASHの病態を修飾していると考えられる.また,肝内鉄過剰の原因として消化管からの鉄吸収が亢進していることが推測された.その機序は,十二指腸粘膜での鉄吸収分子であるDMT1,DcytbおよびHephaestinの発現が上昇していることによると考えられた.一方,消化管からの鉄吸収が亢進する機序の一つとしてHepcidin産生が低下している可能性を想定し検討したが,その発現量は鉄過剰状態を反映し増加していた. NASHに対する除鉄療法(瀉血+低鉄栄養療法)は,血清ALT値およびIV型コラーゲン値を有意に改善させ,かつ組織学的改善も得られ有効な治療であることを明らかにした.さらに,治療前増加していた酸化的DNA障害のマーカーの一つである8-OH-dG量が除鉄療法を48週間施行することにより低下することも見出した.したがって,除鉄療法により酸化ストレスを軽減することにより,肝炎を沈静化させるのみならず,将来的にはNASHからの肝発癌率を低下させうる可能性が示唆された.

TGF-βは上皮細胞に対して増殖抑制に作用するが,多くのがん細胞ではTGF-β刺激に不応性であることからそのシグナル伝達経路に何らかの異常が存在すると推定されている.しかし、過半数の癌ではTGF-β刺激に対して不応となる機序は不明であり.未知のシグナル伝達分子が想定されている.申請者らはこれまでTGF-βのシグナル伝達機構において癌抑制遺伝子p53が重要な役割を担っていることを見い出し報告してきた.即ち,TGF-β receptor(TβR)にrecruitされたprotein phosphatase 2A(PP2A)のBα subunitが,TβRのserine/threonine kinaseによりリン酸化/活性化され,p53のセリン376の脱リン酸化を惹起Ep53の核移行を促進して細胞周期停止シグナルを伝達していることをを見い出している.以上の結果より,TGF-βによる増殖抑制シグナルからの逸脱を生じる原因として、癌において高頻度に認められるp53の変異・欠損も関与していると推察される。しかし、このTGFβ-PP2A-p53シグナルにおいてkey moleculeであるPP2A-Bαの癌における異常は報告されていない。平成18年度ではin vitroの系として,PP2A-Bαの変異欠損の有無を樹立細胞株を用いて探索し,癌との関係について基礎的検討を行った.その結果,PP2A-Bαをノックダウンした細胞では足場非依存性増殖,浸潤能の充進さらには腫瘍原性を獲得することを確認した.さらに,平成18-19年度にかけてPP2A-Bαノックアウトマウスを作製したが,胎生致死でありこの分子は発生に極めて重要な分子であることが判明した.現在,発生毅階におけるPP2A-Bαの役割を分析すると共に,conditional Koマウスを作製し更なる検討を行っている.

神経線維腫1型患者で悪性末梢神経鞘腫瘍（MPNST）を3回外科的に切除したFFPE組織から、神経線維腫とMPNSTのレーザーマイクロダイセクションにより腫瘍切片を切り出し、タンパク質を抽出した。これら3病変に共通でMPNSTに有意に増加していたタンパク質は321種、神経線維腫に有意に増加していたタンパク質は18種であった。増加したタンパク質群のKEGG pathway解析で、MPNSTではタンパク質合成・プロセッシング・分解過程の全てが活性化され、特にプロテアソーム関連タンパクが多く増加(Fold Enrichment:7.93)していた。抗体入手が可能なプロテアソームサブユニット20S proteasome β2、PSMD2は免疫組織染色にてMPNSTで発現が亢進していた。このことからプロテアソーム選択的阻害剤がMPNSTに対する治療効果を示す可能性が示唆された。そこでMPNST細胞株sNF96.2,SCC94,SCC24で、プロテアソーム選択的阻害剤の暴露を行い、細胞株での増殖抑制を確認した。

In this study, we investigated the BH3 profiling and drug delivery system to clarify the anticancer drug susceptibility by a new reagent to soft tissue sarcoma using liposome bound with Fluorodeoxyglucose (FDG). This clarified the therapeutic and synergistic effects of various anticancer agents in soft tissue sarcoma.

The failure of normal hematopoiesis is observed in myeloid neoplasms. However, the precise mechanisms governing replacement of normal hematopoietic stem cells in their niche by myeloid neoplasm stem cells have not been clarified yet. Primary acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS) cells induced aberrant expression of multiple hematopoietic factors including JAG1, SCF and ANGPT1 in mesenchymal stem cells. Importantly, transfer of myeloid neoplasm-derived extracellular vesicles reduced the hematopoietic-supportive capacity of mesenchymal stem cells. Analysis of exosomal microRNA indicated that several species including miR-7977 from acute myeloid leukemia cells were higher than those from normal CD34+ cells. Remarkably, the copy number of miR-7977 in BM interstitial fluid was elevated in not only AML, but also MDS, as compared with control bone marrow. Thus, miR-7977 in extracellular vesicles may be a critical factor that induces failure of normal hematopoiesis.
