Roles of gap junctions in glucose transport from glucose transporter 1-positive to -negative cells in the lateral wall of the rat cochlea

Despite the importance of glucose metabolism for auditory function, the mechanisms of glucose transport in the cochlea are not completely understood. We hypothesized that gap junctions mediate intercellular glucose transport in the cochlea in cooperation with facilitative glucose transporter 1 (GLUT1). Immunohistochemistry showed that GLUT1 and the tight junction protein occludin were expressed in blood vessels, and GLUT1, the gap junction proteins connexin26 and connexin30, and occludin were also present in strial basal cells in the lateral wall of the rat cochlea. Gap junctions were found among not only these GLUT1-positive strial basal cells but also GLUT1-negative fibrocytes in the spiral ligaments and strial intermediate cells. Glucose imaging using 6-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-6-deoxyglucose (6-NBDG, MW 342) together with Evans Blue Albumin (EBA, MW 68,000) showed that 6-NBDG was rapidly distributed throughout the stria vascularis and spiral ligament, whereas EBA was localized only in the vessels. The gap junctional uncouplers heptanol and carbenoxolone inhibited the distribution of 6-NBDG in the spiral ligament without decreasing the fluorescence of EBA in the blood vessels. These findings suggest that gap junctions mediate glucose transport from GLUT1-positive cells (strial basal cells) to GLUT1-negative cells (fibrocytes in the spiral ligament and strial intermediate cells) in the cochlea.

Treatment of cells with pervanadate or vanadate induces the phosphorylation of caveolin-1 and its internalization from the cell surface, but the intracellular fate of caveolae has not been fully elucidated. In the present study, we examined the fate of endocytosed caveolae in human umbilical vein endothelial cells and mouse endothelial KOP2.16 cells. The localization of internalized caveolae and their relationship with the endosomes were examined by immunofluorescence microscopy as well as by immunoprecipitation and chasing of biotinylated transferrin. In untreated cells, caveolin-1 was mostly confined to the cell surface. When cells were treated with either pervanadate for 30 min or vanadate for 3 h, many caveolin-1-labeled vesicles were formed inside the cells, some of which were colocalized with Rab5 or Rab4. The internalized caveolin-1 was colocalized with the endocytosed transferrin in the Rab5-, Rab4- or early endosome antigen-1-labeled compartment where caveolin-1 was phosphorylated. It then moved to the Rab11-associated compartment. Immunogold electron microscopy revealed that internalized caveolin-1 colocalized with Rab5 or Rab4 in vesicles larger than caveolae. These results suggest that the internalized caveolae interact with early endosomes.

Water channel aquaporin 5 (AQP5) is present in the apical membrane of the salivary gland acinar cells. We examined changes of AQP5-distribution during the fusion process of secretory granule membranes into the apical membrane and subsequent recovery process in the mouse parotid gland by administering isoproterenol (IPR) in vivo. We performed immunoperoxidase, immunofluorescence and immunoelectron microscopy. In the basal state, AQP5 was localized mainly in the apical membrane of the acinar cell. It was also present in the basolateral membrane to a lesser extent. When IPR was administered to mice, dot-like, vesicle-like and vacuole-like labeling for AQP5 was seen in the subapical regions by light microscopy. By immunoelectron microscopy, AQP5 was localized at both the apical and basolateral plasma membranes in the basal state. At 5 and 30 min after the IPR-administration, acinar lumen became enlarged and small invaginations formed by fusion of secretory granules were seen. AQP5 was positive along the apical plasma membrane and its small invaginations. At 60 min, large invaginations of the lumen were formed. AQP5 remained positive in the membrane of these large invaginations. At 6 h, large invaginations disappeared and AQP5 was localized in the apical plasma membrane. AQP5 was restricted to plasma membranes and continuous invaginations formed by the exocytosis of secretory granules. AQP5 was not detected in the cytoplasm. These observations show that AQP5 does not seem to be endocytosed during the membrane recycling process following the exocytosis.

Gap junctions mediate glucose transport between GLUT1-positive and -negative cells in the spiral limbus of the rat cochlea

To elucidate the role of the spiral limbus in glucose transport in the cochlea, we analyzed the expression and localization of GLUT1, connexin26, connexin30, and occludin in the spiral limbus of the rat cochlea. GLUT1 and occludin were detected in blood vessels. GLUT1, connexin26, connexin30, and occludin were also expressed in fibrocytes just basal to the supralimbal lining cells. Connexin26 and connexin30 were present among not only these GLUT1-positive fibrocytes but also GLUT1-negative fibrocytes. In vivo glucose imaging using 6-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl) amino]-6-deoxyglucose ( 6- NBDG, MW 342) together with Evans Blue Albumin (EBA, MW 68,000) showed that 6- NBDG was rapidly distributed throughout the spiral limbus, whereas EBA was localized only in the vessels. Moreover, the gap junctional uncoupler heptanol inhibited the distribution of 6- NBDG. These findings suggest that gap junctions play an important role in glucose transport in the spiral limbus, i.e., that gap junctions mediate glucose transport from GLUT1-positive fibrocytes to GLUT1-negative fibrocytes in the spiral limbus.

Apical localization of sodium-dependent glucose transporter SGLT1 is maintained by cholesterol and microtubules

A GFP-labeled sodium-dependent glucose transporter SGLT1 (SGLT-GFP) was transfected into MDCK cells. SGLT-GFP was localized at the apical membrane in confluent cells. When cellular cholesterol was depleted by methyl-beta-cyclodextrin (M beta CD) treatment, the localization of SGLT-GFP gradually switched from apical to whole plasma membrane. Time-lapse microscopy revealed that the effect of M beta CD appeared within 30 min, and that the transition of SGLT-GFP to the whole plasma membrane was completed within 2 hr after the administration. Immunofluorescence microscopy revealed that the tight junction framework remained steady during this process. The effect of M beta CD on SGLT-GFP localization was counterbalanced by the addition of cholesterol into the culture medium. Disruption of microtubules by colcemid also perturbed SGLT-GFP localization. SGLT-GFP localized to the whole plasma membrane by colcemid treatment, and apical localization was restored within 1 hr after removal of colcemid. Inhibition of protein synthesis by cycloheximide had no effect on the transition of SGLT-GFP induced by the M beta CD or colcemid. These results indicated that the apical localization of SGLT-GFP is maintained by cellular cholesterol and microtubules, possibly with an apical recycling machinery.

Differential regulation of AQP2 trafficking in endosomes by microtubules and actin filaments

Vasopressin-induced trafficking of aquaporin-2 (AQP2) water channels in kidney collecting duct cells is critical to regulate the urine concentration. To better understand the mechanism of subcellular trafficking of AQP2, we examined MDCK cells expressing AQP2 as a model. We first performed double-immunolabeling of AQP2 with endosomal marker proteins, and showed that AQP2 is stored at a Rab11-positive subapical compartment. After the translocation to the plasma membrane, AQP2 was endocytosed to EEA1-positive early endosomes, and then transferred back to the original Rab11-positive compartment. When Rab11 was depleted by RNA interference, retention of AQP2 at the subapical storage compartment was impaired. We next examined the role of cytoskeleton in the AQP2 trafficking and localization. By the treatment with microtubule-disrupting agent such as nocodazole or colcemid, the distribution of AQP2 storage compartment was altered. The disruption of actin filaments with cytochalasin D or latrunculin B induced the accumulation of AQP2 in EEA1-positive early endosomes. Altogether, our data suggest that Rab11 and microtubules maintain the proper distribution of the subapical AQP2 storage compartment, and actin filaments regulate the trafficking of AQP2 from early endosomes to the storage compartment.

Aquaporin-2 (AQP2) is one of the membrane water channel proteins expressed in principal cells of the kidney collecting ducts. In the basal state, AQP2 resides in the storage vesicles localized in the subapical cytoplasm. Upon stimulation with vasopressin, AQP2 is translocated to the apical plasma membrane by the exocytic fusion of the storage vesicles with the apical membrane. This translocation enables the transepithelial reabsorption of water from the lumen to the interstitium via AQP2 at the apical membrane and AQP3/AQP4 at the basolateral membrane. AQP2-storage vesicles are distinct from the endoplasmic reticulum, Golgi apparatus, trans-Golgi network, and lysosomes. The early endosomal marker EEA1 is colocalized with some of AQP2 vesicles. Further analyses in Madin-Darby canine kidney (MDCK) cells transfected with AQP2 revealed that subapical Rab11-positive/EEA1-negative smaller vesicles constitute part of the AQP2 storage vesicles for the translocation to the apical membrane. Termination of stimulation results in the retrieval of AQP2 to the larger EEA1-positive early endosomal compartment. AQP2 is then transferred to the subapical storage compartment in a PI3-kinase-dependent manner. GLUT4 is an isoform of glucose transporters whose localization is also regulated by vesicular trafficking induced by insulin stimulation. Comparison of the intracellular localization of AQP2 with GLUT4 suggests distinct regulation of AQP2 trafficking.

Aquaporin-2 (AQP2) is one of the water-channel proteins expressed in principal cells of kidney collecting ducts, where it is stored in the intracellular compartment. Previous studies have demonstrated that AQP2 vesicles constitute a distinct intracellular compartment partially overlapping with early endosomes. In this report, we performed in vitro experiments using the renal epithelia] cell line, Madin-Darby canine,kidney (MDCK) cells, stably expressing AQP2 (MDCK-hAQP2). In nonpolarized cells, AQP2 vesicles were scattered in the cytoplasm and did not colocalize with Golgi 58K or TGN38. Small portions of AQP2 vesicles were positive for the lysosome marker cathepsin D. An early endosome antigen (EEA1) localized around AQP2 vesicles in close proximity, suggesting involvement of the endosomal system in the trafficking of AQP2. AQP2 vesicles are distinct from other recycling molecules, such as glucose transporter 4 (GLUT4) and endocytosed transferrin. In polarized MDCK-hAQP2 cells, AQP2 vesicles were localized in the subapical recycling compartment and distinct from the Golgi apparatus, trans-Golgi network, lysosome, and early endosome in the nonstimulated. state. When the cells were treated with forskolin, translocation of AQP2 to the apical membrane was observed. Washout of forskolin induced retrieval of AQP2 into the cytoplasm, and AQP2 was transiently colocalized with EEA1-positive endosomes. Then,. AQP2 moved from EEA1-positive endosomes to the subapical AQP2-storage compartment, which is sensitive to wortmannin and LY294002. These results suggest that AQP2 resides in a recycling compartment at the apical side in polarized MDCK-hAQP2 cells, and its retrieval uses the apical endosomal system and the phosphatidylinositol 3-kinase-dependent pathway.

Expression of genes followed by the examination of the localization of their products by histochemical methods is one of important and powerful approaches to elucidate the role of genes of interest. We show here a simple and versatile electroporation method for the introduction of plasmids into cells cultured on coverslips for histochemical examination. Electroporation apparatus was assembled using a 35-mm plastic dish and aluminum wire. The apparatus was placed over the coverslip and plasmid DNA was applied onto it. Green fluorescent protein (GFP) was used as a reporter of transfection. Under the appropriate settings of voltage, duration, and pulse numbers, successful transfection was easily achieved in 3T3-L1 fibroblasts, HeLa cells, Cos7 cells, and MDCK cells. The electroporation method described here is simple, easy, rapid, and economical, and may be best suited for the screening of many genes for their histochemical examination.

Immunolabeling of cultured cell lines of epithelial origin is often performed to study cell polarity. To explore the localization of cellular molecules in detail, we devised an efficient and convenient method to prepare semithin and ultrathin cryosections of cells cultured on permeable support. Multiple specimens were plied up to obtain many vertical images of cells in a single section. Freezing them in cooled n-hexane resulted in clear and homogeneous frozen specimens, from which semithin and ultrathin cryosections of epitherial cells on permeable support were easily obtained. With this method, it is possible to prepare many cryosections of good quality for histochemical examination of vertical images of cells.

Introduction and expression of glucose transporters in pancreatic acinar cells by in vivo electroporation

cDNAs encoding facilitated-diffusion glucose transporters GULT1, GULT3, GULT4, and GULT5 were introduced into the rat pancreas by in-vivo electroporation method, and their expression and localization in pancreatic acinar cells were examined immunohistochemically. GLUT1 was localized at the basolateral membrane, whereas GLUT3 and GLUT5 were at the apical membrane. Restriction of GLUT3 and GLUT5 to the apical membrane makes a marked contrast to their localization to the entire plasma membrane when expressed in hepatocytes in situ. Such differential localization may be due to differential apical targeting mechanism: direct targeting in acinar cells and indirect transcytotic delivery in hepatocytes in situ. GLUT4 was present in the membrane of zymogen secretory granules in the cytoplasm in acinar cells. This observation suggests that GLUT4 is segregated to the regulated secretory pathway. Expression of glucose transporters in situ is a useful method in analyzing the targeting mechanism of membrane proteins in cells of tissues in situ.

Internalization of caveolae structure on the cell surface is promoted by oxidant stress or inhibitor of phosphatase (Aoki et al., Exp Cell Res 253, 629, 1999). The internalization is accompanied by phosphorylation of caveolin-1 , a marker protein of caveolae. Little is known about the significance of this internalization. Recently we showed that phosphorylated caveolin-1 forms a complex with Rab5 protein. The existence of Rab5-caveolin-1 complex suggests that internalized caveolac could be translocated to endosomes. To see whether early endosome marker protein EEA1 is related to caveolae, we examined internalized caveolae by immunofluorescence microscopy, cryoultrathin immunogold electron microscopy, immunoprecipitation. Immunofluorescence examination revealed that internalized caveolae colocalized with EEA1 in many area. By immunoprecipitation, both caveolin-1 antibody and phosphorylated caveolin-1 antibody precipitated EEA1 protein. When caveolae were internalized, EEA1 antibody effectively precipitated many phosphorylated caveolin-1 . On ultrathin cryosections of cultured endothelial cells and caveolae-rich fractions obtained by sucrose density-centrifugatioin method, we confirmed that EEA1 colocalized with caveolae structure. These results show a close relationship between EEA1 and caveolae structure.

Rabbits were immunized with partial peptides corresponding to dog Na +-coupled myoinositol cotransporter (SMIT) and rat water channel aquaporin 3 (AQP3). Immunoblot analysis and immunofluorescence microscopy revealed that the antibodies corresponding to SMIT and to AQP3 both detected AQP3 protein in the kidney. Neither of them, on the other hand, detected SMIT protein. Immunogen peptides of both SMIT and AQP3 contain the identical sequence "KEQI", whereas other amino acids are different from each other. Immunohistochemical preabsorption studies demonstrated that the antibodies corresponding to SMIT mainly recognized the sequence "KEQI" as an epitope. These observations demonstrate that the utmost care should be taken in using anti-peptide antibodies.

Background. Fructose is a nutrient as well as a potent agent for the formation of advanced glycation end product in diabetes. GLUT5 is a facilitated-diffusion fructose transporter expressed in the small intestine and kidney. Previous reports on the localization of GLUT5 by in situ hybridization and immunohistochemistry were controversial. Methods. The expression of GLUT5 was checked by reverse transcription-polymerase chain reaction and Southern blotting and immunoblotting analyses. Localization of GLUT5 was visualized by high-resolution immunofluorescence and immunogold electron microscopy. Results. We were able to confirm the expression of GLUT5 in the kidney. GLUT5 was predominantly present in the outer stripe of the outer medulla, where it was localized in the S3 proximal tubule cells. Double labeling with phalloidin showed that GLUT5 was localized in the brush border of the S3 proximal tubule cells. Ultrastructural examination revealed that GLUT5 was present along the plasma membrane of the apical microvilli. Conclusion. GLUT5 is present at the apical plasma membrane of S3 proximal tubule cells and may serve as the transporter of fructose.

Peptide transporter-1 is a H+/peptide cotransporter responsible for the uptake of small peptides and peptide-like drugs, and is present in the absorptive epithelial cells of the villi in the small intestine (duodenum, jejunum, and ileum). It has been localized to the apical microvillous plasma membrane of the absorptive epithelial cells of the rat small intestine using the immunogold electron microscopic technique. Digital image analysis of the jejunum revealed that the transporter protein was abundant at the tip of the villus and that the amount decreased from the tip of the villus to its base. The effect of dietary administration of amino acids and starvation on the expression of PepT1 in the jejunum was examined by immunoblotting and image analysis of immunofluorescence. Starvation markedly increased the amount of peptide transporter present, whereas dietary administration of amino acids reduced it. The gradient of the transporter protein along the crypt-villus axis was maintained under either condition. These observations show that it is specific to the microvillous plasma membrane and that its expression is regulated by the nutritional condition.

Aquaporins (AQPs) are membrane water channel proteins expressed in various tissues in the body. We surveyed the immunolocalization of AQP3, an isoform of the AQP family, in rat epithelial tissues. AQP3 was localized to many epithelial cells in the urinary, digestive, and respiratory tracts and in the skin. In the urinary tract, AQP3 was present at transitional epithelia. In the digestive tract, abundant AQP3 was found in the stratified epithelia in the upper part, from the oral cavity to the forestomach, and in the simple and stratified epithelia in the lower part, from the distal colon to the anal canal. In the respiratory tract, AQP3 was present in the pseudostratified ciliated epithelia from the nasal cavity to the intrapulmonary bronchi. In the skin, AQP3 was present in the epidermis. Interestingly, AQP3 was present at the basal aspects of the epithelia: in the basolateral membranes in the simple epithelia and in the multilayered epithelia at plasma membranes of the basal to intermediate cells. During development of the skin, AQP3 expression commenced late in fetal life. Because these AQP3-positive epithelia have a common feature, i.e., they are exposed to an environment of possible water loss, we propose that AQP3 could serve as a water channel to provide these epithelial cells with water from the subepithelial side to protect them against dehydration.

Phosphatidylinositol 3-kinase (PI3-kinase) is a heterodimeric enzyme composed of a catalytic subunit of 110 kDa and an adaptor regulatory subunit. We investigated the presence and localization of five isoforms of the regulatory subunits, p55α, p55γ, p85α, p85β, and p50α, in the rat brain. In situ hybridization histochemistry using isoform-specific cRNA probes revealed that all five isoforms were expressed in the neurons of the brain. Interestingly, most neuronal cells including Purkinje cells in the cerebellum and pyramidal cells in the cerebrum expressed all five isoforms. Immuno-histochemical staining also showed the localization of p55α, p55γ, p85α, and p50α in the neuronal cells in the brain. Expression of multiple isoforms in neurons suggests that they may play important roles in signal transduction in the brain.

In vivo electro-transfection efficiency and manner of transferred gene expression were investigated by fluorescence microscopic image analysis. Green fluorescent protein (GFP) gene was used as the genetic marker, Electroporation was carried out on the liver of live rats by use of disk electrodes mounted in the tips of tweezers, which were directly pressed onto the surface of a liver lobe in situ. Electroporation with eight electric pulses of 50 ms in duration at 50 V gave a good efficiency of transfection as judged by the induced GFP expression. Bright fluorescence of GFP appeared as dots, which were scattered around the area damaged by electroporation. The transfection efficiency increased as the amount of injected DNA was increased. The results indicate that the amount of induced gene expression can be controlled. Estimation of the efficiency of electro-gene transfer using the fluorescence of GFP and digital analysis of microscopic images was useful to determine the optimum conditions for local gene therapy in tissues and organs.

Twenty-one nucleic acid-specific fluorochromes were examined for use in cell nuclear counter-staining of specimens observed under a laser confocal microscope equipped with a Krypton/argon laser tube. Six green, three red, and two far-red fluorochromes gave sufficient fluorescence. Among them, SYBR Green I, Pico Green, SYTOX Green, YO-PRO-3, and TO-PRO-3 specifically stained the cell nuclear DNA. Other fluorochromes except POPO-3 stained the cell nuclear DNA and cc-stained the cytoplasmic RNA. Conversely, POPO-3 specifically stained the cytoplasmic and nucleolar RNA. Anti-bleaching reagents (PPDA and DABCO) effectively prevented the bleaching of SYBR Green I, Pico Green, YO-PRO-8, and TO-PRO-3, whereas DABCO accelerated the photobleaching of SYTOX Green. Taking into account the characteristics of these fluorochromes, SYBR Green I, YO-PRO-3, and TO-PRO-3 were best suited for cell nuclear counter-staining as green, red, and far-red fluorochromes, respectively. Especially, TO-PRO-3 seems to be the most useful because its far-red fluorescence enables three-color fluorescence staining by combination with immunofluorescence labeling with fluorescein and rhodamine derivatives.

We have investigated the subcellular distribution of GLUT4 by immunofluorescence microscopy after transfection with wild-type or mutant dynamin cDNA into Chinese hamster ovary cells expressing insulin receptor and GLUT4. In the basal state, GLUT4 was distributed exclusively within the cells in the cells overexpressing wild-type dynamin (CHO(IR-GLUT4)-WT) but was located at the cell surface in the cells overexpressing mutant dynamin (CHO(IR-GLUT4)-K44E). Insulin induced subcellular shift of GLUT4 to the cell surface in CHO(IR-GLUT4)-WT cells but had little effect in CHO(IR-GLUT4)-K44E cells. When insulin-stimulated cells were treated with wortmannin, GLUT4 was redistributed within the cells in CHO(IR-GLUT4)-WT cell, whereas it remained at the cell surface in CHO(IR-GLUT4)-K44E cell. These results suggest that dynamin is a regulatory GTPase in endocytosis of GLUT4.

Twenty nucleic acid-specific fluorescent dyes were screened for use in nuclear staining in laser confocal microscopy. Eleven of them, BO-PRO-1, YO-PRO-1, SYBR Green I, PicoGreen, SYTOX Green, TO-PRO-1, POPO-3, propidium iodide, YO-PRO-3, TOTO-3, and TO-PRO-3 gave sufficient fluorescence by in-gel assay. They were also tested for the nuclear staining in cultured cells. When glycerol was used as a mounting medium, it enhanced the fluorescence intensity and retarded the bleaching in most of nucleic acid staining dyes. Among green fluorochromes excited at 488 nm, YO-PRO-1, SYTOX Green, and SYBR Green I were suitable for nuclear DNA staining. YO-PRO-1 showed the strongest fluorescent intensity, but it weakly co-stained the cytoplasmic and nucleolar RNAs. SYTOX Green strongly and specifically stained the nuclear DNA, Among red fluorochromes excited at 568 nm, propidium iodide stained nuclear DNA with simultaneous staining of RNA. YO-PRO-3 strongly and specifically stained DNA but bleached relatively rapidly. TO-PRO-3 excited at 647 nm was specific for DNA, but its fluorescence was bleached rapidly. Selection of nuclear DNA staining dyes should be done by considering the fluorescence intensity, DNA specificity, and bleaching characteristics.

Localization of facilitated-diffusion glucose transporter GLUT1 was examined in the rat mammary gland by immunofluorescence microscopy and immunoperoxidase electron microscopy. Secretory epithelial cells of acini were enriched with GLUT1, which was localized to the basolateral plasma membrane. Apical plasma membrane and cytoplasmic organelles including the Golgi apparatus were negative for GLUT1. In addition to acini. abundant GLUT1 was found in the perineurium nerve fiber bundles. which is a site of blood-nerve barriers. GLUT1 in the basolateral membrane of secretory epithelial cells seems to serve as machinery to take up glucose into the cell for milk synthesis and secretion.

We examined five nucleic acid binding fluorescent dyes, propidium iodide, SYBR Green I, YO-PRO-1, TOTO-3, and TO-PRO-3, for nuclear DNA staining, visualized by fluorescence and laser confocal microscopy. The optimal concentration, co-staining of RNA, and bleaching speeds were examined. SYBR Green I and TO-PRO-3 almost preferentially stained the nuclear DNA, and the other dyes co-stained the cytoplasmic RNA. RNAse treatment completely prevented the cytoplasmic RNA staining. In conventional fluorescence microscopy, these dyes can be used in combination with fluorescence-labeled antibodies. Among the dyes tested, TOTO-3 and TO-PRO-3 stained the DNAs with far-red fluorescence under red excitation. Under Kr/Ar- laser illumination, TOTO-3 and TO-PRO-3 were best suited as the nuclear staining dyes in the specimens immunolabeled with fluorescein and rhodamine (or Texas red).

SGLT1, an isoform of Na+-dependent glucose transporters, is localized at the apical plasma membrane in the epithelial cells of the small intestine and the kidney. In the present study we examined its location in SGLT1 cDNA-transfected MDCK cells, which form an epithelial sheet connected by tight junctions in culture. Formation of tight junctions was monitored by staining for occludin, an integral tight junction protein. In the cells demarcated by an uninterrupted occluding meshwork, SGLT1 was specifically localized at the apical plasma membrane, showing that SGLT1 has a signal to accomplish this restricted localization. In the cells with little or no occludin accumulation in the tight junction, however, SGLT1 was present along the entire aspect of the plasma membrane. Similar distribution of SGLT1 was observed in the cells as long as the occludin meshwork remained incomplete. These observations suggest that apical localization of SGLT1 occurs upon the completion of the uninterrupted meshwork of tight junctions.

Elevation of intra-cellular organelle DNA levels due to preferential synthesis of organelle DNAs before cell multiplication is generally observed in meristematic cells of higher plants, such as those in apical meristems and cultured cells. We analyzed the physiological significance of this phenomena using cultured tobacco cell BY-2 as a model system for cell multiplication in plants. Cultured tobacco cell BY-2 multiplied approximately 50-fold in a week in normal culture medium. However, when nalidixic acid, which inhibits prokaryotic DNA gyrase, was added to the culture, synthesis of organelle DNA was selectively inhibited and cell multiplication was arrested. This indicates that the synthesis of organelle DNAs is a prerequisite for cell multiplication. To quantitatively clarify the effect of the elevation of organelle DNA levels on the capacity for cell proliferation, cells with various organelle DNA levels were prepared by preculturing cells for various lengths of time in culture medium which contained aphidicolin, a specific inhibitor of eukaryotic DNA polymerase α, and then transferring them to culture medium which contained nalidixic acid. Their growth rates in the absence of further organelle DNA synthesis were monitored. A correlation was observed between intra-cellular organelle DNA levels and the capacity of the cells to proliferate, indicating that intra-cellular organelle DNA levels limited the capacity for cell proliferation.
