Localization of facilitated-diffusion glucose transporter GLUT1 was examined in the rat mammary gland by immunofluorescence microscopy and immunoperoxidase electron microscopy. Secretory epithelial cells of acini were enriched with GLUT1, which was localized to the basolateral plasma membrane. Apical plasma membrane and cytoplasmic organelles including the Golgi apparatus were negative for GLUT1. In addition to acini. abundant GLUT1 was found in the perineurium nerve fiber bundles. which is a site of blood-nerve barriers. GLUT1 in the basolateral membrane of secretory epithelial cells seems to serve as machinery to take up glucose into the cell for milk synthesis and secretion.

Nuclear staining for laser confocal microscopy

Twenty nucleic acid-specific fluorescent dyes were screened for use in nuclear staining in laser confocal microscopy. Eleven of them, BO-PRO-1, YO-PRO-1, SYBR Green I, Pico Green, SYTOX Green, TO-PRO-1, POPO3, propidium iodide, YO-PRO-3, TOTO-3, and TO-PRO-3 gave sufficient fluorescence by ingel assay. They were also tested for the nuclear staining in cultured cells. When glyoerol was used as a mounting medium, it enhanced the fluorescence intensity and retarded the bleaching in most of nucleic acid staining dyes. Among green fluorochromes excited at 488nm, YO-PRO-1, SYTOX Green, and SYBR Green I were suitable for nuclear DNA staining. YO-PRO-1 showed the strongest fluorescent intensity, but it weakly costained the cytoplasmic and nucleolar RNAs. SYTOX Green strongly and specifically stained the nuclear DNA. Among red fluorochromes excited at 568nm, propidium iodide stained nuclear DNA with simultaneous staining of RNA. YO-PRO-3 strongly and specifically stained DNA but bleached relatively rapidly. TOPRO-3 excited at 647 nm was specific for DNA, but its fluorescence was bleached rapidly. Selection of nuclear DNA staining dyes should be done by considering the fluorescence intensity, DNA specificity, and bleaching characteristics.

We examined five nucleic acid binding fluorescent dyes, propidium iodide, SYBR Green I, YO-PRO-1, TOTO-3, and TO-PRO-3, for nuclear DNA staining, visualized by fluorescence and laser confocal microscopy. The optimal concentration, co-staining of RNA, and bleaching speeds were examined. SYBR Green I and TO-PRO-3 almost preferentially stained the nuclear DNA, and the other dyes co-stained the cytoplasmic RNA. RNAse treatment completely prevented the cytoplasmic RNA staining. In conventional fluorescence microscopy, these dyes can be used in combination with fluorescence-labeled antibodies. Among the dyes tested, TOTO-3 and TO-PRO-3 stained the DNAs with far-red fluorescence under red excitation. Under Kr/Ar- laser illumination, TOTO-3 and TO-PRO-3 were best suited as the nuclear staining dyes in the specimens immunolabeled with fluorescein and rhodamine (or Texas red).

SGLT1, an isoform of Na+-dependent glucose transporters, is localized at the apical plasma membrane in the epithelial cells of the small intestine and the kidney. In the present study we examined its location in SGLT1 cDNA-transfected MDCK cells, which form an epithelial sheet connected by tight junctions in culture. Formation of tight junctions was monitored by staining for occludin, an integral tight junction protein. In the cells demarcated by an uninterrupted occluding meshwork, SGLT1 was specifically localized at the apical plasma membrane, showing that SGLT1 has a signal to accomplish this restricted localization. In the cells with little or no occludin accumulation in the tight junction, however, SGLT1 was present along the entire aspect of the plasma membrane. Similar distribution of SGLT1 was observed in the cells as long as the occludin meshwork remained incomplete. These observations suggest that apical localization of SGLT1 occurs upon the completion of the uninterrupted meshwork of tight junctions.

Elevation of intra-cellular organelle DNA levels due to preferential synthesis of organelle DNAs before cell multiplication is generally observed in meristematic cells of higher plants, such as those in apical meristems and cultured cells. We analyzed the physiological significance of this phenomena using cultured tobacco cell BY-2 as a model system for cell multiplication in plants. Cultured tobacco cell BY-2 multiplied approximately 50-fold in a week in normal culture medium. However, when nalidixic acid, which inhibits prokaryotic DNA gyrase, was added to the culture, synthesis of organelle DNA was selectively inhibited and cell multiplication was arrested. This indicates that the synthesis of organelle DNAs is a prerequisite for cell multiplication. To quantitatively clarify the effect of the elevation of organelle DNA levels on the capacity for cell proliferation, cells with various organelle DNA levels were prepared by preculturing cells for various lengths of time in culture medium which contained aphidicolin, a specific inhibitor of eukaryotic DNA polymerase α, and then transferring them to culture medium which contained nalidixic acid. Their growth rates in the absence of further organelle DNA synthesis were monitored. A correlation was observed between intra-cellular organelle DNA levels and the capacity of the cells to proliferate, indicating that intra-cellular organelle DNA levels limited the capacity for cell proliferation.
